Quick answer
All of this is to be carried on in constant, Spirit-led prayer, watchful and persevering, including intercession for all of God's people. It matters because the armor is worn in dependence on God, expressed through prayer.
Overview
Prayer is not a separate piece of armor so much as the atmosphere in which the whole battle is fought, marked by 'all' prayer, 'all' times, 'all' perseverance, and 'all' the saints. 'In the Spirit' shows that true prayer is enabled and directed by the Holy Spirit. The call to watchfulness and persistence reflects the seriousness of the conflict and the believer's ongoing dependence on God, with concern that reaches beyond self to the whole church.
